The Nigerian opened the scoring for the Turkish champions with a penalty kick in the fourth minute after a handball from a Kayserispor defender.
The hosts, however, drew level 10 minutes later through Cameroon international Stephen Bahoken, whose thumping header from a corner kick beat goalkeeper Fernando Muslera.
Galatasaray got back in the lead through Baris Alper Yilmaz in the 29th minute, with the Turkish winger ensuring his side heads into the break with the lead.
Yunus Akgun doubled the visitors’ lead with a top-class goal six minutes into the second half, having been teed up by Dries Mertens.
Akgun then provided for Osimhen, who dinked the ball over the on-rushing goalkeeper to complete his brace, taking his season tally to 12 goals and five assists in 15 matches across all competitions.
Galatasaray got their fifth goal of the tie through Yilmaz in the 87th minute with Akgun serving as provider once more.
The win helped the champions maintain the top spot in the Turkish Super Lig standings, opening an eight-point lead over Fenerbahce, who were held to a 1-1 draw at Eyupspor on Friday.
